ELISA 酶結合物的制備:免疫酶技術(immunoenzymatic technique)又稱酶免疫測定法(enzyme immunoassay,EIA),是繼免疫熒光技術和放射免疫技術之后發展起來的又一種免疫標記技術。它是把抗原抗體的特異性反應和酶的高效催化作用相結合而建立的。該技術通過化學方法將酶與抗體或抗抗體結合,形成酶標記物,這些酶標記物仍保持免疫學活性和酶的活性,然后將它與相應的抗體或抗原起反應,形成酶標記復合物,結合在免疫復合物上的酶,在遇到相應的底物時,則催化底物產生水解、氧化或還原等反應,而生成可溶性或不溶性有色物質。然后根據顯色的深淺來反映待測樣品中抗原或抗體的含量,如生成的有色物質為可溶性,則可用肉眼比色或酶標測定儀測定光密度值(OD)定性或定量;如生成的有色物質為不溶性沉淀物,同時又是電子致密物質,則可用光學顯微鏡或電子顯微鏡識別和定位。因此,免疫酶技術是一項定位、定性和定量(敏感度可達每毫升ng~pg水平)的綜合技術。常用的酶是辣根過氧化物酶(horseradish peroxidase,HRP)和堿性磷酸酶(alkaline phosphatase,AKP)。
